چکیده

The preferential absorption of near infrared light by blood makes photoacoustic imaging well suited to visualising vascular structures in soft tissue. In addition, the spectroscopic specificity of tissue chromophores can be exploited by acquiring images at multiple excitation wavelengths. This allows the quantification of endogenous chromophores, such as oxyand deoxyhaemoglobin, and hence blood oxygenation, and the detection of exogenous chromophores, such as functionalised contrast agents. More importantly, this approach has the potential to visualise the spatial distribution of low concentrations of functionalised contrast agents against the strong background absorption of the endogenous chromophores. This has a large number of applications in the life sciences. One example is the structural and functional phenotyping of transgenic mice for the study of the genetic origins of vascular malformations, such as heart defects. In this study, photoacoustic images of mouse embryos have been acquired to study the development of the vasculature following specific genetic knockouts.

In vivo longitudinal photoacoustic imaging of subcutaneous tumours in mice

Photoacoustic tomography can provide high resolution 3D images of vascular networks, making it well suited to characterising the development of tumour vasculature and its response to treatment. In this study, photoacoustic images to depths of up to 9 mm were obtained using an all optical ultrasound detection scheme. Two type of colorectal tumours (LS174T and SW1222) implanted subcutaneously in ...

B-Mode Photoacoustic Imaging using Linear Array: Numerical Study for Forward-Backward Minimum Variance Beamformer Combined with Delay-Multiply-and-Sum

Photoacoustic imaging (PAI) is a promising medical imaging modality which provides the resolution of Ultrasound (US) and the contrast of Optical imaging modalities. One of the most important challenges in PAI is image formation, especially in the case that a linear-array US transducer is used for data acquisition. This is due to the fact that in the linear-array scenario, there is only 60 degre...

Acoustic attenuation compensation in photoacoustic tomography: Application to high-resolution 3D imaging of vascular networks in mice

The reconstruction algorithms commonly used in photoacoustic tomography do not account for the effects of acoustic attenuation on the measured time-domain signals. For experimental measurements made in biological tissue, acoustic attenuation causes the high frequency components of the generated ultrasound signals to be significantly reduced. When this signal loss is neglected, it manifests as a...

In vivo 3-dimensional photoacoustic imaging of the renal vasculature in preclinical rodent models.

Non-invasive imaging of the kidney vasculature in preclinical murine models is important for studying renal development, diseases and evaluating new therapies, but is challenging to achieve using existing imaging modalities. Photoacoustic imaging is a promising new technique that is particularly well suited to visualising the vasculature and could provide an alternative to existing preclinical ...

Evaluation of a 99mTc-tricine Vascular Disrupting Agent as an In-vivo Imaging in 4T1 Mouse Breast Tumor Model

Colchicine as a vascular disrupting agent creates microtubule destabilization whichinduces vessel blockage and consequently cell death. Accordingly, colchicines and itsanalogues radiolabeled with 99mTc may have potential for visualization of tumor. In this work, deacetylcolchicine a colchicine analogue was labeled with 99mTc via tricine as a coligandand characterized for its tumor targeting prop...
